Does anyone know if it is possible to change some of the "defaults" for the ServiceNow alert destination in Classic Synthetic? Specifically the Assignment group and Assigned to fields?

No, the Classic Service Now integration is not configurable. This functionality is available on Dynatrace Synthetic but not on Classic.
Support can provide an alternative approach with PowerShell Script that polls the Alert API and can be modified to meet your needs, please contact support if you wish to go that route.
